Molecular Structure and Sequence of Aminkovs
1. molecular structure:
The molecular structure of the products can be divided into three parts, namely the N-terminal peptide of GLP-1, the C-terminal peptide of GCG and the long-chain peptide connecting the two. The specific structure is shown in the figure below:
It can be seen below that the molecular structure is similar to that of GLP-1 and GCG, but the long-chain peptides linked between them are different. In this long-chain peptide, there are many unnatural amino acids (such as Arg, Pro, Gln, etc.) to increase its stability and effectiveness. In addition, some special structural units such as N-ethylmalonyl-L-arginine-N’-tert-butoxycarbonyl (MPA) and 3-methoxytyrosine (3- methoxytyrosine) were introduced into the molecular structure to further enhance its biological activity.
2. amino acid sequence:
GLP-1+GIP consists of a polypeptide sequence of up to 42 amino acids. Its specific amino acid sequence is:
Tyr-Gly-Gly-Phe-Leu-Gly-Arg-Pro-Arg-Arg-Gln(1)-Lys-Glu-Val-Lys-Asp-(Mpa)-(N-ethylproline)-({{20} }methoxytyrosine) -(N-ethylproline)-(D-amino acid)-(N-methylglycine).
Among them, Tyr, Gly, Phe, Leu, Lys, Glu, Val, Asp, etc. are the standard 20 types of natural amino acids, while Arg, Pro, Gln and the unnatural amino acids in the back brackets are introduced into the sequence to improve stability and efficiency.
